Substratum-attached material ("footpads") remaining after treatment of A. coffeaeformis with EGTA. The footpads are soluble in hot bicarbonate solution and are stained here with Concanavalin A conjugated to fluorescein isothiocyanate (100 μgmL-1, Sigma Chemical Co.). Red autofluorescence of the diatom chloroplast is seen above the footpads when diatoms were not removed from the substratum.
